Spectrin is a cytoskeletal protein that lines the intracellular side of the plasma membrane of many cell types in pentagonal or hexagonal arrangements, forming a scaffolding and playing an important role in maintenance of plasma membrane integrity and cytoskeletal structure. In certain types of brain injury such as diffuse axonal injury, spectrin is irreversibly cleaved by the proteolytic enzyme calpain, destroying the cytosketelon. Spectrin cleavage causes the membrane to form blebs and ultimately to be degraded, usually leading to cell death. The finding that mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC) has resulted in considerable interest in the understanding of the mechanism of DNA mismatch repair. Initially, inherited mutations in the MSH2 and MLH1 homologs of the bacterial DNA mismatch repair genes mutS and mutL were demonstrated at high frequency in HNPCC and were shown to be associated with microsatellite instability. A member of the mismatch repair family, GTBP (also designated MSH6), is an MSH2-related protein that binds to DNA containing G/T mismatches. Findings suggest that the mismatch-binding factor in human cells is composed of a heterodimer of GTBP and MSH2. MITF (microphthalmia transcription factor) is a basic helix-loop-helix-leucine-zipper (bHLH-Zip) transcription factor that regulates the development and survival of melanocytes and retinal pigment epithelium, and also is involved in transcription of pigmentation enzyme genes such as tyrosinase TRP1 and TRP2. MITF has been shown to be phosphorylated by MAP kinase in response to c-kit activation, resulting in upregulation of MITF transcriptional activity. Mutations of the MITF gene are associated with the autosomal dominant hereditary deafness and pigmentation condition, Waardenburg Syndrome type 2A. Multiple isoforms of MITF exist, including MITF-A, MITF-B, MITF-C, MITF-H, and MITF-M, which differ in the amino-terminal domain and in their expression patterns. Napsin is a pepsin-like aspartic proteinase connected with maturation of surfactant protein B. There are two closely related napsins, napsin A and napsin B. Napsin A is expressed as a single chain protein. Immunohistochemical studies revealed high expression levels of napsin A in human lung and kidney but low expression in spleen. Napsin A is expressed in type II pneumocytes and in adenocarcinomas of lung. The high specificity expression of napsin A in adenocarcinomas of lung is useful to distinguish primary lung adenocarcinomas from adenocarcinomas of other organs. This MAb recognizes the peptide core of gastric mucin M1 (recently identified as Mucin 5AC). Its epitope is located in the N-terminal cysteine rich part of the peptide core of MUC5AC, which is heavily glycosylated. Its epitope is destroyed by beta-mercaptoethanol but not by periodate treatment. MAb 2-11M1 reacts with the protein backbone exclusively; it only reacts with fully deglycosylated MUC5AC. Therefore, the material under test should also be fully deglycosylated. This can be achieved with standard periodate oxidation method. The success of the deglycosylation can be checked with routine PAS (Periodic Acid Shiff) staining. After deglycosylation, the preparation should no longer be stainable with PAS reagent. Only then 2-11M1 will react should MUC5AC be present. The mini-chromosome maintenance (MCM) family of proteins, including MCM2, MCM3, MCM6 (Cdc21), MCM5 (Cdc46), MCM6 (Mis5) and MCM7 (Cdc47), are regulators of DNA replication that act to ensure replication occurs only once in the cell cycle. Expression of MCM proteins increases during cell growth, peaking at G1/S phase. The MCM proteins each contain an ATP-binding motif, which is predicted to mediate ATP-dependent opening of double-stranded DNA. This antibody recognizes an epitope encoded by exon v9 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA. Complement C1q B Chain (C1qb) is a subunit of C1q. C1q is composed of 6 polypeptide chains each of C1qa, C1qb, and C1qc. C1q is a component of the classical complement pathway, and thereby plays an important role in the immune response. Each C1q subunit contains an N-terminal collagen-like region and a C-terminal C1q globular domain. The presence of receptors for C1q on effector cells modulates its activity, which may be antibody-dependent or independent. Macrophages are the primary source of C1q, while anti-inflammatory drugs as well as cytokines differentially regulate expression of the mRNA as well as the protein.
